Magigen Bst DNA Polymerase, Large Fragment, is the portion of the Bacillus stearothermophilus DNA Polymerase protein that contains the

5´ → 3´ polymerase activity and strong strand displacement activity, but lacks 5´ →3´ exonuclease activity. Bst could be used for isothermal amplification reactions, such as LAMP (Loop-mediatedisothermal amplification)，RCA (Rolling-circle amplification), etc.

● How to use Bst polymerase

Taking LAMP amplification as an example:

Configure the reaction system according to the following table:

Incubate at a constant temperature of 60 ° C-65 ° C for 45-60 minutes.

● Cautions

a. The combination and amount of amplification primers can be optimized and adjusted according to the actual situation. It is recommended that the reaction time should not exceed 60min.

b. If fluorescence detection is used, additional nucleic acid dye shall be added.

c. If the additional physiological saline content added to the overall system exceeds 10 μl, Amplification will not proceed normally, and it is necessary to increase the usage of Bst enzyme.
