Intended use

Magigen Mouse Tail Genotyping Kit is used for genotyping of transgenic animals such as mice. This mouse genotyping kit is optimized to fast isolate mouse tail DNA for genotyping. It provides a rapid, ultra-high sensitive, highly efficient and low-cost method in the DNA extraction and PCR amplification from mouse tail, ear, and other tissue for mouse genotyping.

No need nucleic acid extraction: run PCR amplification directly after animal tissue lysed.

Simple and rapid operation: DNA extraction and amplification in as little as 1 hour. Magigen genotyping kit should be the first choice for high-throughput genotyping in laboratory.

The product can be electrophoresed directly. It allows direct loading of PCR products onto agarose gels for analysis without adding any buffer after PCR amplification.

Detection principle

Magigen™ Mouse Tail Genotyping Kit includes a proprietary lysis buffer and PCR reaction system. The cells of transgenic mice, such as mouse tail, and other animal tissues, can be lysed directly under the action of the lysis buffer. It is no need for mechanical break-up, organic extraction, column purification, and DNA precipitation to obtain genomic DNA. Mouse genomic DNA can be extracted in 15 minutes.

Magigen Mouse Genotyping kit uses the improved MagicScript HST enzyme (HotStart Taq DNA polymerase) for PCR amplification. It is only needed a little number of samples to complete the amplification and analysis of the target gene when using this kit for PCR detection.

Operation procedure

Take the mouse tail specimen as an example.

1.     Snip the mouse tail

1.1   Collect tiny small pieces (about 3- 5mm) of mouse tail into a micro-tube fitting to your PCR machine holes.

1.2   Lyse the tissue. 18μL Lysis Buffer A diluting with162μL ddH2O, add it into the tube, and heat it in a water bath at 95℃ for 10 min. Make sure the tissue is immersed in the solution instead of sticking on tube wall. Make sure the cap is securely sealed. It would result in failure if tube leaking during heating.

1.3   Stop lysis. Adjust the PCR environment, add 20μL lysis buffer B, mixed well, put the tube into PCR thermocycler, centrifuge at 4℃ for 3~5min.

1.4   Amplification. Take 0.5 ~ 2μL supernatant for PCR amplification.
